    CHCT Leadership and Organization

    Director: Leslie E. Silberstein, M.D.

    Dr. Leslie Silberstein received his Baccalaureate and M.D. Degrees from the University of Leiden, the Netherlands, and had post-graduate training in Hematology/Oncology and Transfusion Medicine at Tufts-New England Medical Center in Boston, MA. He is currently a Professor of Pathology at Harvard Medical School and Director of the Joint Program in Transfusion Medicine at Children's Hospital, Boston, Brigham and Women’s Hospital, and the Dana-Farber Cancer Institute. In addition, he is also a Senior Investigator at the Immune Disease Institute.

    Co-Directors:Jerome Ritz, M.D., and David Scadden, M.D.

    Dr. Jerome Ritz received his Baccalaureate degree from Northwestern University in Evanston, Illinois and his M.D. from Chicago Medical School, in Chicago, Illinois. He had post-graduate training in Internal Medicine at the University of Wisconsin Hospitals in Madison, Wisconsin and at Beth Israel Hospital, in Boston, Massachusetts. He is currently a Professor of Medicine at Harvard Medical School and the Medical Director of the Connell and O’Reilly Families Cell Manipulation Core Facility at Dana-Farber Cancer Institute.
    Dr. David Scadden received his Baccalaureate degree from Bucknell University in Lewisburg, Pennsylvania and his M.D. from Case Western Reserve in Cleveland, Ohio. He had post-graduate training in Medicine at Harvard Medical School, Brigham and Women's Hospital, and Dana-Farber Cancer Institute, in Boston, Massachusetts. He is currently Professor of Medicine at Harvard Medical School, the Co-Director of the Harvard Stem Cell Institute, Director of the Massachusetts General Hospital (MGH) Center for Regenerative Medicine, and Chief of Hematologic Malignancies Program at the MGH Cancer Center.

    Translational Cell Therapy Laboratory

    The Translational Cell Therapy Laboratory (TransLab) serves as a two-way transfer between basic science and patient care with the goal of developing better and newer cell-based therapies. A team of experienced personnel address the issues involved in cell therapy development: scaling up techniques to meet the volumes associated with clinical trials, developing closed systems of culture whenever possible, developing release criteria and potency assays for the cellular products generated and writing Standard Operating Procedures (SOPs). Once the SOPs are developed and written, they are transferred to the appropriate staff in the Cell Manipulation Core Facility at the DFCI for validation of each SOP.

    Regulatory Core

    The purpose of the Regulatory Core of CHCT is to provide consultation service to scientific and clinical investigators on regulatory issues throughout the development phase of the studies. The Assistant Medical Director, Dr. Grace Kao, the Quality Assurance Manager, Olive Sturtevant, and Research and Development Manager, Dr. Elena Spanjaard, of the Cell Manipulation Core Facility at Dana-Farber Cancer Institute will be the major contributors for this core facility. Beside working closely together with the staffs at the Translational laboratory to develop procedures adherent to current Good Manufacture Practice (cGMP) and Good Tissue Practice (cGTP), the Regulatory Core will also provide manufacture information to the investigators for submission of Investigational New Drug (IND) applications to the Food and Drug Administration (FDA).

    Flow Cytometry Core

    All the projects conducted at the CHCT TransLab depend on the use of flow cytometry. The Flow Cytometry Core is equipped with 2 flow cytometers: a 2 laser-6 colors BD FACS Canto and a high-speed cell sorter and analyzer (Dako Cytomation MoFlo). The MoFlo is a 3 laser system configured with a 488nm, 633nm and UV lasers. A variety of techniques are routinely performed: Immunophenotyping (surface and intracellular antigens), apoptosis analysis (Annexin V, PI), cell cycle analysis (PI, 7-AAD) and proliferation (CFSE) and cell sorting.
